Binding of EGF o r TGF alpha to EGFR activates tyrosine kinase activity of the recept o r (2). Phosph o r ylation of Tyr 992, Tyr 1068 a n d Tyr 1086 is required f o r conf o r mational change in the C-terminal of EGFR (3). Autophosph o r ylation of Tyr 992 c r e a t e s a binding site f o r the phospholipase C-gamma (PLC-gamma) SH2 domain, inducing downstream signaling (4).
